YPD Growth Medium

YPD is used for the growth of Saccharomyces cerevisiae. It is a nutrient-rich medium containing dextrose, peptone, and yeast extract. Dextrose is the energy source. Yeast extract and peptone provide carbon, nitrogen, minerals, vitamins, trace elements, and other essential growth nutrients that stimulate yeast growth.
One sachet is dissolved in 500ml of water and sterilized in an autoclave. Homebrewers can use a microwave to disinfect the contents in a borosilicate bottle with the lid slightly loose. Avoid superheating. Remove from the microwave, place aluminum foil around the bottleneck. When the bottle has cooled down to approximately 70℃, you can screw on the cap (not too tightly).
YPD medium is commonly used in microbiology in combination with agar to create agar plates, a useful tool for investigating contamination, for starting a culture, isolation, growth experiments, cell counting, and as an indicator of phenotypic characteristics (what an organism does visually differently from another organism).
Equipment for making agar growth medium in a petri dish, also called an agar plate:
  • Borosilicate bottle 500ml
  • YPD
  • Agar
  • Petri dish

Method - enough for pouring 20 agar plates with 9 cm diameter

  1. Weigh 10 g agar (2%) together with 25g YPD medium and let swell for 15 minutes in 500 mL room temperature tap water in a borosilicate bottle.

  2. Disinfect (boil) the mixture in a microwave oven, avoiding superheating - screw the lid on so that it gives a little.

  3. Gently mix the solution, avoiding air bubbles, and cool the bottle to approximately 60 ◦C.

  4. Distribute the mixture evenly into 20 agar plates. 25 mL per plate, or to a height of just under 0.5 cm

    When pouring the mixture into the dishes, it is enough to fill the dish halfway, then carefully place the lid over it and swirl the dish to distribute the solution evenly.

  5. Let the dishes solidify overnight. The next day, they are ready for use.
